Paper Review: Identification of an emerging cucumber virus in Taiwan using Oxford nanopore sequencing technology………..Daniel Bojorquez Flores………..Cucurbit viruses represent a serious problem that affects crop production in Taiwan, negatively impacting yield and fruit quality. In June 2020, cucumber plants cultivated in greenhouses in Xizhou Township of Changhua County showed severe symptoms of leaf mosaic and fruit deformation. Samples from these symptomatic plants were collected to diagnose the already known and commonly reported cucurbit virus, however, the diagnosis results were negative. This suggested that the symptoms could be associated with an emerging virus.
To identify the causal agent, the authors isolated the viral particles from infected plants and mechanically inoculated quinoa leaves with them. The leaves were ground and prepared for the transmission electron microscopy (TEM) to observe the viral particles. Additionally, the double stranded RNA was extracted from infected quinoa leaves and sequenced using the Oxford nanopore sequencer to obtain the viral genome.
Furthermore, bioinformatic analysis of the resulting sequences revealed that the virus was closely related to Cucumber Bulgarian latent virus (CBLV). Finally, the authors developed specific diagnostic tools that allow the virus detection and could facilitate future monitoring and management of cucurbit crops in Taiwan.
